If the enzyme is not able to repair the damage, the cell can die. The objective of this project is to understand how these enzymes function. Specifically, the project is focused on the xeroderma pigmentosum group C (XP-C) gene. It encodes the ATPase subunit of the DNA excision repair (ER) enzyme XP-C endonuclease, and in the absence of the XP-C gene function, XP-C endonuclease and ER are incapable of repairing DNA. There are at least four known forms of XP-C (xeroderma pigmentosum complementation groups C, D, G, and E). These can be divided into two types: Type A proteins (XP-C and XP-C group D) have defects in both ER and XP-C endonuclease; type B proteins (XP-G and XP-E) have defects in XP-C endonuclease. The type A group have reduced but not abolished DNA repair activity, whereas the type B group have no DNA repair activity. In addition to XP-C endonuclease, the gene encodes a protein with homology to a number of bacterial ATP-dependent helicases. This protein appears to be required for DNA replication. The long term objective of the project is to understand the mechanism of DNA repair by XP-C endonuclease. The first specific aim is to study the structure of the type B and type A proteins. The type B group contains very few amino acid differences from the type A group. Therefore, a combination of peptide mapping and mass spectrometry will be used to determine the structure of the type B proteins.
